Macromolecular crowding develops heterogeneous environments of gene expression in picoliter droplets

چکیده

Drawing of the PDMS-based microfluidic chip. Two inlets including one for the oil and one for the in vitro transcription and translation mix (IVTT). Droplets are generated using PDMS based microfluidic chips, the IVTT mixture is pinched off at regular intervals by oil generating monodisperse water in oil emulsions. The droplets are stored in the storage chamber and can be monitored over time with fluorescence microscopy. Plasmid map of pRSET-CFP/ YFP (see Methods section for more details).
